Brandon Hospital was opened on January 22nd, 1928 during a pivotal period in Poplar Bluff's medical history. According to historical documentation from Poplar Bluff Regional Medical Center, Brandon Hospital is described as "one of the first hospitals in Poplar Bluff," reflecting its important role in establishing professional healthcare infrastructure in the community. Records indicate the hospital previously operated at 216 S. Main according to a 1923 article.

The hospital building became a recognizable landmark in the city and was frequently featured on postcards and promotional materials of the era. These postcards, which survive in historical collections today, provide valuable visual documentation of early 20th-century medical architecture in rural Missouri.
